摘要:【目的】基于免移蟲育王技術探究幼蟲信息素酯類的三種組合3E(1.0%甲基亞油酸酯、16.0%甲基亞麻酸酯、35.0%甲基油酸酯)、4E(4.5%甲基棕櫚酸酯、1.0%甲基亞油酸酯、16.0%甲基亞麻酸酯、35.0%甲基油酸酯)和10E(4.5%甲基棕櫚酸酯、2.5%甲基硬脂酸、35.0%甲基油酸酯、1.0%甲基亞油酸酯、16.0%甲基亞麻酸酯、3.5%乙基棕櫚酸酯、1.5%乙基硬脂酸酯、18.0%乙基油酸酯、0.5%乙基亞油酸酯、17.5%乙基亞麻酸酯)在育王中對中華蜜蜂Apis cerana cerana蜂王質量的影響?!痉椒ā坷弥腥A蜜蜂免移蟲育王生產器培育蜂王,在幼蟲60-64h時向王臺內注入1μL配制好的酯類混和物(濃度梯度分別為0%、0.1%、1.0%、10.0%),待蜂王出房后測定蜂王初生重、胸重、胸寬以及單側卵巢管數(shù)量,并利用熒光定量PCR測定蜂王卵巢卵黃蛋白原基因Vitellogenin(Vg),昆蟲儲存蛋白基因hexamerin70b (hex70b)和hexamerin110(hex110)的表達量。【結果】免移蟲育王過程中添加3E,不同濃度處理蜂王的初生重均顯著增加,但蜂王胸寬和胸重均無顯著變化;0.1%和1.0%濃度處理蜂王卵巢管數(shù)顯著增加;不同濃度處理Vg和hex70b的表達量均無顯著變化,但1.0%和10.0%濃度處理蜂王卵巢的hex110表達量顯著升高。4E試驗組,只有10.0%濃度處理蜂王的初生重和卵巢管數(shù)顯著上升,其它濃度處理對蜂王的個體指標均無顯著影響;0.1%和1.0%濃度處理Vg的表達量顯著上升,10.0%處理下hex70b的表達量顯著增加,1.0%和10.0%濃度處理hex110表達量也顯著上升。10E試驗組,各個濃度處理蜂王的初生重、Vg和hex110的表達量均顯著下降,但蜂王的卵巢管數(shù)和胸部指標無顯著變化,10.0%的濃度處理hex70b的表達量也顯著降低。【結論】育王過程中添加幼蟲信息素酯類對中華蜜蜂育王質量存在影響,1.0%的3E顯著增加蜂王的初生重、卵巢管數(shù)和hex110的表達量;10.0%的4E也顯著增加蜂王的初生重和卵巢管數(shù),同時蜂王卵巢hex70b和hex110的表達水平顯著上升。

Abstract:【Aim】The objective of this study is to investigate the effects of 3E( 1.0%methyl linoleate, 16.0%methyl linolenate, 35.0%methyl oleate), 4E(4.5%methyl palmitate, 1.0%methyl linoleate, 16.0%methyl linolenate, 35.0%methyl oleate) and 10E(4.5% methyl palmitate, 2.5% methyl stearate, 35.0% methyl oleate, 1.0% methyl linoleate, 16.0% methyl linolenate, 3.5% ethyl palmitate, 1.5% ethyl stearate, 18.0% ethyl oleate, 0.5% ethyl linoleate, 17.5% ethyl linolenate) on the quality of queen reared by queen-rearing technology without larvae-grafting. 【Methods】Queen-rearing technology without larvae-grafting was employed to rear queen of Apis cerana cerana. 1μL of combination of brood pheromone esters with concentration gradient of 0, 0.1%, 1.0% and 10.0% was added into the queen cells respectively using a micro sampling syringe injector when queen larvae was 60-63h old. The weight, number of ovariole tube in one side, the weight and width of thorax of newly-emerged queens were tested and compared. And gene expression of Vitellogenin(Vg) , hexamerin70b(hex70b) and hexamerin110(hex110) was quantified by qRT-PCR.【Results】The results showed that the newly-emerged weight was increased significantly (P<0.05) in all concentration treatments of 3E, but no significant effects were found in the thorax index and ovarioles number of reared queens compared with control (P>0.05). The queens have higher expression of hex110 gene in 1.0% and 10.0% concentration of (P<0.05), while in Vg and hex70b genes there was no significant difference among the experimental groups of 3E. The weight and ovarioles number was significantly increased when the queen was reared with 10.0% of 4E (P<0.05). expression of Vg in queen’s ovary was significantly higher in 0.1% and 1.0% treatment of 4E (P<0.05), while higher expression of hex70b existed in 10.0% (P<0.05), and hex110 in 1.0% and 10.0% treatment of 4E (P<0.05). In 10E, the newly-emerged weight, expression of Vg and hex110 gene were all lower in all treatments than those in control, while in the ovarioles number and thorax index no significant difference were found. expression of hex70b reduced significantly only in 10% treatment of 10E (P<0.05).【Conclusion】Combinations of brood pheromone esters applied in queen-rearing process could affect the queen’s quality, 1% treatment of 3E significantly increased the newly-emerged weight and the ovarioles number, and increased the expression of hex110 gene significantly in queen ovary. In 10% treatment of 4E, the newly-emerged weight, the ovarioles number and the expression of hex70b and hex110 of queen were all increased significantly.
